如何选购ELISA试剂

ELISA试剂盒在实验室已 经相当普及,绝大多数ELISA试剂盒都是用于检测未知样本中抗原的浓度。用试剂盒当然比自己慢慢摸条件快得多,ELISA试剂盒不仅可以让实验更便利, 往往还更加。现在市面上的ELISA试剂盒既有国产也有进口,数量繁多令人目不暇接,怎样才能选出zui适用的产品呢?首先当然得根据我们所要检测的分子 进行检测,除此以外也还有一些需要加以考量的因素,本文就来为您介绍一二。

1. 实验类型

ELISA试剂盒有多种类型,不过它们都有一个共同特点,就是进行抗原捕获。一般捕获形 式包括将抗原吸附到微孔板或者用微孔板上的抗体进行捕获。In-cell ELISA和酶联免疫斑点ELISPOT是两种特殊的类型,In-cell ELISA需要将微孔板中培养的细胞进行固定和通透化,然后再用抗体原位检测。ELISPOT有点像蛋白印迹Western blot,先在带膜的微孔板中培养细胞,然后在膜上检测细胞分泌的抗原形成斑点。此外,我们还需要根据自身需要选择96孔板或是384孔板进行ELISA 实验。

通常人们使用双抗夹心法进行ELISA实验,双抗夹心法中抗原被夹在捕获抗体和检测抗体 之间,这种方法既灵敏又有效,受到了许多研究者的青睐。不过有时候双抗夹心法并不是*选择,例如有时候抗原特别小让两个大抗体难以附着,又或者抗体只有 一个抗体结合位点。在上述情况下,竞争性ELISA就更为适用,这种方法是采用带标记的纯化抗原与样本中无标记的抗原进行竞争,以结合捕获抗体。

2. 抗体类型

单克隆抗体和多克隆抗体都能够用于ELISA,实际上有时候二者结合效果更好。例如,对于双抗夹心法而言,用多抗进行捕获而单抗进行检测有时更有帮助。多抗能够确保捕获所有抗原,随后单抗再特异性检测拥有特定抗原表位的抗原。

双抗夹心法ELISA中捕获抗体与检测抗体(不论多抗还是单抗)的配对很有讲究。我们不希望捕获抗体与检测抗体竞争抗原上的同一位点,为此我们就需要确 保捕获抗体和检测抗体所识别的抗原表位不存在重叠。如果捕获抗体和检测抗体之间发生了冲突,就会对实验结果产生较大影响。要避免这一问题,我们可以选择购 买“matched pair”的捕获/检测抗体对,这些打包在一起的抗体是商家经过验证才推荐的好组合。

3. 交叉反应

如果抗体间发生冲突或交叉反应就会影响整个实验的特异性。其中抗体来源所带来的兼容性就 是交叉反应的一个因素。尽管现在购买源自动物的抗体越来越容易,我们仍有必要确认一下是否会产生交叉反应的问题。在用双抗夹心法进行ELISA检测 时,检测用的二抗必须特异性针对检测一抗,而不能与捕获抗体起反应。如果检测用二抗与捕获抗体结合,实验特异性就会大打折扣。通常我们选用源自不同宿主的 捕获抗体和检测一抗,来避免上述问题。

与此同时,了解试剂盒中提供的buffer也是有必要的(例如washing buffer和blocking buffer),以免这些buffer含有可能影响抗原抗体相互作用的组分,使检测结果收到影响。

4. 检测方式

如今检测ELISA有许多途径,我们可以根据自己的偏好进行选择。一般ELISA检测的 是酶促反应的可溶性产物,抗体上结合有相应的酶(例如通常使用的辣根过氧化物酶HRP),而反应体系中添加有相应的底物(如3,3-二氨基联苯胺 DAB)。这种酶促反应生成具有特征性的颜色,可以通过分光光度计进行检测,碱性磷酸酶AP也是一种常用酶。酶可以结合在一抗上,也可以结合在二抗上,还 可以使用链霉亲和素标记的酶与亲和素标记的一抗结合。此外ELISA的结果检测还包括放射性检测、荧光检测、化学发光或显色法检测等。

使用微孔阵列的说明

  • 1. 用一把镊子提起微孔阵列的边缘,将刀片从载玻片上剥离下来。在与微孔阵列一起使用时,请务必戴手套,因为皮肤上的污染物(例如灰尘,油脂等)可能会对阵列的粘合性能产生不利影响。

  • 2. 通过从一侧边缘放下插入物,将微细胞阵列小心地放置在成像培养皿中,以确保在培养皿表面和微细胞阵列之间没有残留空气。如果气泡被困住,请取下插入件,然后重试。如果仍然继续捕获气泡,请将一到两滴乙醇放在培养皿上,然后将插入物放下。请注意,如果您使用乙醇帮助将插入物放下,请将培养皿放在通风橱中至少60分钟以使乙醇蒸发,否则添加培养基时插入物将不会粘在细胞培养皿上。

  • 注意:要检查阵列下是否有残留的气泡,请将培养皿上下颠倒并翻到底面。

  • 3. 微孔阵列插入物以真空密封包装提供;但是,我们建议您执行以下灭菌步骤。吸取70-100%乙醇在微孔阵列的顶部,直到包括插入物的培养皿的底面被覆盖。使用1,000μL移液管循环培养皿中的乙醇,以帮助去除可能滞留在孔中的任何气泡。后,使用移液器从培养皿中抽出几乎所有乙醇,确保阵列始终被液体覆盖,并作为废物丢弃。

  • 注意:微孔阵列非常疏水,如果*吸去乙醇漂洗液,则向阵列中添加温热介质会导致气泡滞留在孔中。

  • 4. 慢慢地将热介质添加到文化器具的一个角落,直到几乎装满为止。

  • 5. 从培养皿的对角抽出培养基/乙醇混合物,注意微孔阵列始终被液体覆盖。

  • 6. 重复步骤4和5至少五次,以确保乙醇被*稀释。
