药物靶向——GalNAc

使用与肝细胞中发现的 Ashwell-Morell 受体(去唾液酸糖蛋白受体或 ASGPR)结合的 N-乙酰半乳糖胺 (GalNAc) 偶联物靶向药物递送治疗有效载荷已成为一种递送方法,尤其是治疗性寡核苷酸。多年来,我们通过合成开发单聚体和多聚体碳水化合物配体(包括 GalNAc),在该领域活跃了很多年,用于包括药物靶向在内的应用。

疫苗开发——多糖化学

细菌是高度糖基化的病原体。糖结合疫苗已被证明是预防传染病的成功策略,预计 2021 年全球肺炎球菌疫苗市场将超过 80 亿美元。作为 CRO,我们一直是各种糖结合疫苗开发项目的重要贡献者包括多价(23 血清型)疫苗的临床前开发,该疫苗可能成为复杂的广谱肺炎球菌疫苗。

癌症——聚糖

碳水化合物-蛋白质(凝集素)相互作用

细胞表面装饰有密集且多样的碳水化合物结构(聚糖),这些结构与锚定在细胞膜中的蛋白质或脂质结合。这些可以统称为糖萼。在无数的功能中,特定的聚糖结构与互补的碳水化合物识别/结合蛋白凝集素结合,以调节细胞生理。有许多种类的凝集素,自由循环或与细胞表面连接,参与许多生物过程。研究较为活跃的两种是唾液酸结合免疫球蛋白型凝集素 (Siglecs) 和 β-半乳糖苷结合凝集素 (Galectins)。几种凝集素,包括去唾液酸糖蛋白受体 (ASGPR),已被 FDA 批准用于靶向药物递送系统。扩大该领域的知识为治疗干预提供了新的机会。凭借 20 年合成多种聚糖系统的经验,我们在这里帮助推进该领域的发现和治疗应用。
