本免疫荧光染色试剂盒(IFK)设计用于确定在冷冻和石蜡包埋的组织制剂中的目标抗原的定位。 0到150个幻灯片。

简介:α2-抗纤溶酶(α2-Antiplasmin, α2-AP)是由肝脏合成分泌的,含有452个氨基酸的单链糖蛋白,分子量为70 000,属于丝氨酸酶抑制物家族成员之一,在人血浆中α2-AP是主要的纤溶酶抑制物,在控制纤溶中起关键作用。

实验:

为此制备α2-AP单克隆抗体,建立ELISA方法用于纤溶系统的监测,为进一步提高抗原的免疫原性,减少抗原用量,延长抗原释放时间,用大分子惰性物质如硝酸纤维素膜(NC膜)作为抗原载体进行脾内包埋免疫。我们将抗原吸附于NC膜上,沿小鼠左肩正中线下胸背部纵形切口1 cm长,逐层入腹,轻轻牵出脾脏,以刀尖沿脾纵轴轻划一小口,置入1个三角形载有α2-AP抗原的NC膜,使之*包埋于脾脏内,观察无出血后回纳脾脏,依次缝合伤口。

间隔3 w后,用同样方法再进行1次,于加强免疫后第3天取小鼠脾细胞与骨髓瘤细胞(5∶1),用PEG-4000常规方法进行融合[2,3],HAT培养基选择培养,当克隆长至1/10孔大小时,采用间接ELISA法筛选分泌抗体阳性杂交瘤细胞株,并采用有限稀释法对阳性杂交瘤细胞进行亚克隆,建立了2株稳定分泌抗-α2-AP单克隆抗体的杂交瘤细胞株,分别命名为3H4和2E3,并用α2-AP采用常规方法免疫家兔,制备抗-α2-AP多克隆抗体血清。

对其中的1株3H4进行研究,其杂交瘤染色体众数为96.37±1.90,分泌的抗体为IgG1亚类,kappa型。ELISA检测腹水效价1∶106,采用饱和硫酸铵盐析法从腹水纯化McAb 3H4,SDS-PAGE检测有较高纯度。3H4抗体对α2-AP标准品为阳性反应,而对ATIII及纤维蛋白原均为阴性,说明对其无交叉反应。此杂交瘤细胞株3 d传1代,体外传代培养6个月以上,ELISA检测抗体滴度无明显降低。采用ELISA法检测正常人血浆α2-AP含量,以纯化的McAb 3H4包板,兔抗α2-AP为多抗,辣根过氧化物酶标记的羊抗兔IgG作为三抗,以ABTS为底物,ELISA法检测了30名正常人枸缘酸钠抗凝血浆(1∶9)α2-AP含量,其结果为(98±1.99) mg/L。

应用范围:由此可见,制备α2-抗纤溶酶单克隆抗体用以检测α2-AP,对于了解肝病、血栓性疾病、DIC等许多疾病纤溶改变有着广泛的应用前景,并可用于溶栓治疗监测[1]

PLA是一种高灵敏度蛋白质检测新技术,通过一对核酸适体探针识别目标蛋白分子。当目标蛋白相互作用时,它们在空间位置上相互接近,并与特异核酸链连接,并经PCR反应扩增特异核酸链,来实现对相互作用的目标蛋白分子检测、定位和定量。

 

该研究结果显示,通过PLA方法可以原位检测胶质瘤内MHCⅡ分子和突变型异柠檬酸脱氢酶1(IDH1R132H)蛋白的相互作用,确定这些蛋白的免疫抗原性和抗原表位。作者认为该检测技术能用于获得在胶质瘤组织中是否存在特异性抗原的必要信息,有助于指导临床上对具有特异性抗原胶质瘤的免疫治疗。

抗原产品是蛋白质和病毒裂解物,可刺激对抗原敏感的所有供体的体外免疫反应。这消除了选择优势肽或蛋白质的可能性,允许您对所有供体使用一种抗原。

大多数美国人口都接种了破伤风类毒素,30-40% 的人口巨细胞病毒 (CMV) 呈阳性,这使得这两种抗原成为召回分析的理想选择。

我们提供各种抗原来刺激细胞,所有抗原均使用我们的 PBMC 进行生物活性测试。我们的三种回忆抗原可用于研究不同类型的免疫记忆:巨细胞病毒 (CMV)、破伤风和 M1。

MANIPULATION
