PiggyBac转座子系统的剪切和粘贴机制

高效的PiggyBac转座子系统使用剪切粘贴机制将DNA从PiggyBac载体转移到基因组中。如果只需要临时的基因组整合，则可以将Excision的PiggyBac转座酶瞬时表达，以无污染地去除插入片段，从而重建原始的基因组序列。

抗原产品是蛋白质和病毒裂解物，可刺激对抗原敏感的所有供体的体外免疫反应。这消除了选择优势肽或蛋白质的可能性，允许您对所有供体使用一种抗原。

大多数美国人口都接种了破伤风类毒素，30-40% 的人口巨细胞病毒 (CMV) 呈阳性，这使得这两种抗原成为召回分析的理想选择。

我们提供各种抗原来刺激细胞，所有抗原均使用我们的 PBMC 进行生物活性测试。我们的三种回忆抗原可用于研究不同类型的免疫记忆：巨细胞病毒 (CMV)、破伤风和 M1。

巨细胞病毒 gH 五聚体复合物

重组人巨细胞病毒五聚体蛋白复合物，由 gH、gL、UL128、UL130 和 UL131A 蛋白组成，在哺乳动物 HEK293 细胞中产生，纯度高于 95%。该复合物已在 ELISA 中显示与我们的重组NRP-2蛋白结合。

SDS-PAGE：考马斯染色的 SDS-PAGE，显示巨细胞病毒 gH 五聚体复合物。

直接ELISA：板用1000ng、100ng、10ng或1ng NRP-2包被并用每孔 500 µg 的 NRP-2、gH 五聚体或 gB huFc-tag 进行探测，然后在室温下封闭 1 小时。在室温下，用每孔 500 µg 的 NRP-2、gH 五聚体或 gB huFc-tag 对板进行探测 2 小时。对于一抗，绵羊抗 gH 五聚体血清以 1:2,000 使用，小鼠抗 His-tag 抗体以 1:1,200 使用（对于 ZIKV-NS1），gB 不使用一抗。二抗 STAR88P 用于 gH 五聚体，浓度为 1:2,000；STAR147P 用于通过 Fc-tag 以 1:2,000 检测 gB huFc-tag，Biorad 105300 用于以 1:2,000 检测 ZIKV-NS1。所有二次孵育均在室温下进行 45 分钟。使用 TMB 膜底物 (KPL) 进行检测 40 分钟。使用 1M HCl 终止反应。测定证实 NRP-2 与 gH 五聚体特异性结合，但不与 gB 或 ZIKV NS1 结合。

  • 重组人巨细胞病毒五聚体蛋白复合物（菌株 VR1814），由蛋白 UL75 (gH)、UL115 (gL)、UL128、UL130 和 UL131A 组成。

背景

hCMV 可导致免疫受损个体（如移植受者和 HIV 患者）的严重发病/死亡，而先天性 hCMV 感染可导致出生缺陷。

五聚体复合物在所有野生型 hCMV 毒株上均有表达，但在 AD169 等实验室毒株中明显缺失。（弗里德等人。）。这严重限制了疫苗的开发。受体介导的病毒进入内皮细胞需要功能性的 gH 五聚体复合物，而这种复合物是感染个体中抗病毒抗体的主要靶标之一。NAC 的新型重组 hCMV gH 复合物将是实现必要的基础和应用研究的关键。此外，gH 复合物有望为新一代体外诊断检测提供基础。

类似天然的五聚体复合物在人体细胞中产生，以确保所有蛋白质都正确折叠并具有其天然糖基化模式。通过质谱确认纯化复合物中所有五种蛋白质的存在。五聚体复合物以即用型液体形式提供，提供 100ug 和 1mg 小瓶大小，有机会批量购买。

NAC 在其 CMV 系列中提供另外两种抗原：一种天然高纯度抗原，对 IgG 和 IgM 均表现出高亲和力，是抗体捕获测定的理想选择，另一种更适合作为血清学对照的标准裂解物。

巨细胞病毒 gH 五聚体复合物作为目录项目出售，仅用于研究用途和体外诊断测定/试剂盒，因此无需事先与 NAC 达成协议。如果产品旨在用作药物或作为生产药物的手段（包括用作免疫原和用于临床试验），则必须事先与 NAC 签订合同协议。
